摘要

目的 基于生物信息学分析EFEMP1在胃癌组织中表达及意义.方法 利用GEO数据库胃癌数据集采用CIBERSORT 和 ESTIMATE 计算方法计算 374 例胃癌病人肿瘤组织 ImmuneScore,StormalScore 和 ESTIMATEScore,通过生存分析,蛋白互作网络(PPI)构建寻找与预后相关的核心基因,并利用TCGA胃癌数据库和免疫组化染色验证核心基因表达.结果 GEO和TCGA数据库中EFEMP1在正常组织和肿瘤组织中表达无差异性,与胃癌的生存时间和临床分期相关(T和Stage).GSEA显示EFEMP1高表达主要富集于钙信号通路、细胞黏附和细胞外基质受体相互作用通路.EFEMP1表达与ITGB1在肿瘤直径、组织学分级、TNM分期均相关(P<0.05).结论 EFEMP1参与胃癌的发生、发展,在侵袭和转移中可能和ITBG1表达存在协同作用.

Abstract

Objectives To analyze the expression and significance of EFEMP1 in gastric cancer tissues based on bioinformatics.Methods CIBERSORT and ESTIMATE methods were used to calculate ImmuneScore,StormalScore and ESTIMATEScore in 374 gastric cancer patients from the GEO gastric cancer dataset.Protein interaction network(PPI)was constructed to find core genes related to prognosis,and the expression of core genes was verified by TCGA gastric cancer database and immunohistochemical staining.Results The expression of EFEMP1 was similar between normal tissues and tumor tissues in GEO and TCGA databases,and was associated with the survival time and clinical stage of gastric cancer(T and Stage).GSEA showed that EFEMP1 was mainly enriched in calcium signaling pathway,cell adhesion and extracellular matrix receptor interaction pathways.EFEMP1 expression was significantly correlated with ITGB1 expression in tumor size,histological grade,and TNM stage(P<0.05).Conclusion EFEMP1 may play a synergistic role with ITBG1 in the invasion and metastasis of gastric cancer.
