Expression of TCF21 in lung cancer and its effect on angiogenesis
Objective To investigate the expression of nuclear transcription factor 21(TCF21)in lung adenocarcinoma and its effect on the angiogenesis.Methods A total of 20 matched fresh lung adenocarcinoma tissue specimens werecollected from the Eighth People's Hospital of Jinan City.Western blot was used to detect the expressionof TCF21 in lung adenocarcinoma and adjacent tissues.Human lung cancer cell line A549 was infected with lentivirus,andTCF21 overexpression stable cell line was constructed,Western blot was used to detect the protein expression levels ofhypoxia-inducing factor-1 α(HIF-1 α)and vascular endothelial growth factor(VEGF)in the control group,no-load groupand overexpression group.The effect of TCF21 on the tube-forming ability of human umbilical vein endothelial cells(HUVEC)was observed by in vitro angiogenesis experiment.Results The expression of TCF21 in fresh lung adenocarcinoma tissue was lower than that in adjacent tissues(P<0.05).After overexpression of TCF21 in lungadenocarcinoma A549 cell line,the protein expression of HIF-1 α and VEGF was significantly decreased compared withthe MOCK group and Vector group in the control,and the tube length and nodes of HUVEC were significantly reduced in tube formation experiments.Conclusion TCF21 is lowly expressed in lung adenocarcinoma tissue,and has theability to inhibit the angiogenesis of lung adenocarcinoma,providing a new potential target for clinical anti-tumor therapy.
