Effect of PRDX2 regulation of HPV16 E6 on activity of cervical cancer Siha cells
Objective To investigate the expression of PRDX2 in cervical cancer tissues and its effect on the malignant biological behavior of cervical cancer Siha cells.Methods The expression of PRDX2 in cervical cancer tissues and adja-cent tissues was detected by immunohistochemistry.The cervical cancer Siha cells were divided into four groups:Siha group,siRNA NC group,siRNA PRDX2 group and siRNA HPV16 E6 group.The proliferation of Siha cells was detected by EDU staining assay.Transwell assay was used to analyze the invasion number of Siha cells.The apoptosis of Siha cells was detected by flow cytometry.The expression level of PRDX2/HPV16 E6 protein in Siha cells was analyzed by Western blot.Results Compared with the adjacent tissues,the expression of PRDX2 was up-regulated in cervical cancer tissues.Com-pared with Siha group and siRNA NC group,the proliferation ability of Siha cells in siRNA PRDX2 and siRNA HPV16 E6 group was decreased(P<0.05).The number of invasive Siha cells in siRNA PRDX2 and siRNA HPV16 E6 groups de creased(P<0.05).The apoptosis rate of Siha cells in siRNA PRDX2 and siRNA HPV16 E6 groups was increased(P<0.05).The expression of HPV16 E6 protein in Siha cells in the siRNA PRDX2 group was down-regulated(P<0.05).Conclusion The expression of PRDX2 is increased in cervical cancer tissues.After PRDX2 expression is inhibited,the proliferation rate and invasion number of Siha cells are decreased,and the apoptosis rate is increased.The mechanism is related to PRDX2 regulating the expression of HPV16 E6 protein.
