GYY4137通过调控糖酵解-细胞焦亡抑制低氧诱导的大鼠肺动脉平滑肌细胞增殖

目的:探讨外源性硫化氢供体GYY4137对低氧(hypoxia)诱导的大鼠肺动脉平滑肌细胞(PASMCs)的作用及其机制。方法:取生长状态良好的大鼠PASMCs,在细胞密度为60%~70%时,饥饿处理24 h。随机将细胞分成4组:正常对照组、正常+GYY4137组、低氧组和低氧+GYY4137组。通过CCK-8检测GYY4137对正常细胞无影响的安全浓度范围,并进一步检测GYY4137抑制低氧诱导的PASMCs增殖的最适浓度;EdU染色法检测各组PASMCs的增殖情况;Western blot检测各组PASMCs中增殖细胞核抗原(PCNA)及糖酵解和细胞焦亡相关蛋白的表达情况;乳酸试剂盒检测各组PASMCs的乳酸含量;免疫荧光法检测各组PASMCs中己糖激酶2(HK2)和核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)蛋白表达情况;ELISA法检测各组PASMCs中白细胞介素1β(IL-1β)和IL-18的含量。结果:GYY4137抑制低氧诱导的PAMSCs的有效浓度为100 µmol/L(P<0。05)。与低氧组相比,低氧+GYY4137组PCNA蛋白表达显著下降(P<0。05),PAMSCs增殖显著减少(P<0。01),HK2和丙酮酸激酶M2(PKM2)蛋白表达均显著下降(P<0。05,P<0。01),丙酮酸脱氢酶(PDH)蛋白表达显著上升(P<0。05),乳酸含量显著降低(P<0。01),细胞焦亡相关蛋白表达显著下降(P<0。05,P<0。01),免疫荧光显示HK2与NLRP3表达显著降低(P<0。01),ELISA试剂盒检测结果显示IL-1β和IL-18含量显著降低(P<0。05,P<0。01)。结论:GYY4137通过调控糖酵解-细胞焦亡抑制低氧诱导的大鼠PASMCs增殖。
GYY4137 inhibits hypoxia-induced proliferation of rat pulmonary ar-tery smooth muscle cells by regulating glycolysis and pyroptosis
AIM:To investigate the effect and mechanism of the exogenous hydrogen sulfide donor GYY4137 on hypoxia-induced proliferation of rat pulmonary artery smooth muscle cells(PASMCs).METHODS:Rat PASMCs in optimal growth condition were used.Once the cell density reached 60%~70%,the cells were serum starved for 24 h.The cells were then randomly divided into four groups:normal group,normal+GYY4137 group,hypoxia group,and hypoxia+GYY4137 group.A CCK-8 assay was used to determine the safe concentration range of GYY4137 that exerted no adverse effects on normal cells,and the optimal concentration of GYY4137 to inhibit hypoxia-induced proliferation of PASMCs was identified.EdU staining was employed to assess PASMCs proliferation in each group.Western blot analysis was conducted to evaluate the expression of proliferating cell nuclear antigen(PCNA)and proteins related to glycolysis and pyroptosis in PASMCs.Lactic acid content was quantified using a lactic acid assay kit.Immunofluorescence was used to detect the pro-tein expression of hexokinase 2(HK2)and nucleotide-binding oligomerization domain-like receptor protein 3(NLRP3)in PASMCs,and the levels of interleukin-1β(IL-1β)and IL-18 in PASMCs were measured using ELISA.RESULTS:The effective concentration of GYY4137 in inhibiting hypoxia-induced viability of PAMSCs was 100 µmol/L(P<0.05).Com-pared with the hypoxia group,the hypoxia+GYY4137 group showed a significant decrease in PCNA protein expression(P<0.05),reduced PAMSCs proliferation(P<0.01),decreased HK2 and pyruvate kinase isozyme type M2(PKM2)protein expression(P<0.05,P<0.01),increased pyruvate dehydrogenase(PDH)protein expression(P<0.05),and reduced lac-tic acid content(P<0.01).Additionally,the expression of pyroptosis-related proteins was significantly decreased(P<0.015,P<0.01),and immunofluorescence revealed a significant decrease in HK2 and NLRP3 expression(P<0.01).ELISA results showed that IL-1β and IL-18 protein levels were significantly decreased(P<0.05,P<0.01).CONCLU-SION:GYY4137 inhibits hypoxia-induced proliferation of rat PASMCs by regulating glycolysis and pyroptosis.
