Analysis of 49 Medicago ruthenica (L.)Trautv.Germplasms Based on AFLP Molecular Markers
The genetic diversity and structure of forty-nine Medicago ruthenica natural accessions from seven provinces and regions in China were analyzed based on amplified fragment length polymor-phisms (AFLP)markers.The results showed that Medicago ruthenica had a high genetic variability and potential as a source of variation for breeding programs.1)Four pairs of AFLP primers combinations were selected.298 visible AFLP bands were observed,of which 21 9 bands were polymorphic,accounting for 73.5%.54.8 polymorphic bands were amplified with a pair of primer in average.2)Percentage of poly-morphic loci of 49 natural accessions was 32.70% in average.The Nei's genetic diversity index was 0.1 15 and the Shannon diversity index was 0.172.A high level of genetic diversity was revealed.3)The genetic variation of Medicago ruthenica accessions in Hebei province was the most abundant,while that in Liaon-ing province was smaller,there were obvious differences in genetic diversity index among geographical groups.4 )The genetic differentiation coefficient among accessions was 0.6436,which showed that 64.36% of the genetic variation originated within accessions;genetic differentiation coefficient among re-gions was 0.2895,which showed that 28.95% of the genetic variation originated among regions.The ge-netic variation among regions was less than that within accessions.5)Cluster analysis and principal compo-nent analysis showed that the genetic similarity coefficient among 49 Medicago ruthenica accessions was 0.5849 ~ 0.9904,the genetic distance was 0.0096 ~ 0.5363,49 accessions were divided into six types, showing obvious regionality.
