Study on the Content of Anthocyanin and the Activities of Related Synthetic Enzymes in Red Stems of Alfalfa
In order to study the reason for the red color of space mutated alfalfa stems,the red stems of space mutated alfalfa and the green stems of unmutated alfalfa(CK)were used as research materials.The anthocyanin components and content in the stems were detected using ultra-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS),and enzyme activity was measured using UV visible spectro-photometry and ELISA kit.The correlation between each anthocyanin glycoside and enzyme activity was ana-lyzed.The results showed that the content of cyanidins,delphinidins,pelargonidin,peonidin and total antho-cyanin in the red stems of alfalfa were significantly higher than those in green stems(P<0.05).Meanwhile,the activities of phenylalanine ammonia-lyase(PAL),cinnamate 4-hydroxylase(C4H),chalcone synthase(CHS),flavonoid-3-hydroxylase(F3H),and glutathione-S-transferase(GST)in the red stems of alfalfa were significantly higher than those in green stems(P<0.05),and the contents of cyanidin,delphinidin,pel-argonidin,peonidin and total anthocyanin were significantly or extremely significantly positively correlated with PAL,C4H,CHS,F3H and GST activities(P<0.05 or P<0.01).In conclusion,the type and con-tent of anthocyanins may be the main reason for the red color of alfalfa stems.The increase in PAL,C4H,CHS,F3H,and GST enzyme activity promoted the transformation of alfalfa stems from green to red.
