Evaluation of Salt Tolerance in Germination and Seedling Stage of 10 Wild Medicago ruthenica L.from Different Provenances
To screen for salt-tolerant Medicago ruthenica L.germplasm resources,10 wild M.ruthenica materials from different provenances were selected as subjects.The germination rate,germination index,ger-mination potential,radicle length,embryo length,fresh weight,and dry weight of M.ruthenica seeds during germination were measured under 150 mmol/L and 200 mmol/L NaCl stress.Under 200 mmol/L NaCl stress,plant height,survival rate,and biomass at seedling stage were measured.Membership function method was used to comprehensively evaluate the salt tolerance at germination and seedling stages.The results showed that all indexes of M.ruthenica seed at germination and seedling stage decreased with the increase of salt con-centration,but the degree of decrease was varied among different materials.The germination rate,germination potential,germination index and vitality index of M.ruthenica seeds were significantly inhibited by salt stress at 150 mmol/L NaCl,but the radicle length and embryo length were less inhibited.Salt stress of 200 mmol/L NaCl severely inhibited seed germination,most of the seeds lost germination ability,some of the germinated seeds could not grow into normal plants,and some seedlings died.The salt tolerance of 10 M.ruthenica materials was different at the germination stage and seedling stage.The results of membership function analy-sis combined with the test indexes of germination and seedling stage showed that the salt tolerance of 10 M.ruthenica materials was MR2>MR3>MR9>MR5>MR4>MR1>MR6>MR7>MR10>MR8.
