Genetic Diversity and Population Structure Analysis of 67 Alfalfa Cultivars Based on SSR Markers
In this study, the genetic diversity and population genetic structure of 67 domestic and for-eign alfalfa cultivars were analyzed based on SSR molecular markers to explored the genetic relationship of alfalfa germplasm resources,in order to provide a theoretical basis for the identification of alfalfa cultivars and breeding of new cultivars. The results showed that 180 polymorphic bands were amplified with 15 pairs of primers, with a polymorphic site percentage of 96.33%.The average values of polymorphism informa-tion content,Shannon's genetic diversity information index and Nei's gene diversity index were 0.73,0.38 and 0.24,indicating the selected primers had high polymorphism. The 67 cultivars were divided into four groups by UPGMA cluster analysis, and there was no significant correlation between the cluster results and the geographical origin of the cultivar. The population genetic structure analysis based on Structure soft-ware divided 67 cultivars into 2 subgroups and 1 mixed population, which was consistent with the results of principal coordinate analysis(PCoA),indicating that most cultivars(85.07%)showed a single genetic background, while the mixed population exhibited gene penetration and had a more complex genetic back-ground.
