基于复方网络药理学和分子对接技术探究吴门通络解毒汤治疗强直性脊柱炎的作用机制

目的 采用网络药理学与分子对接技术探讨吴门通络解毒汤治疗强直性脊柱炎(ankylosing spondylitis,AS)的作用机制.方法 采用TCMSP、TCMID和BATMAN-TCM数据库筛选通络解毒汤中相关有效成分与靶点;采用GeneCards、Drugbank和TTD数据库收集AS相关靶点;使用Venny2.1.0 软件获得通络解毒汤与AS的交集靶点;构建有效成分-交集靶点网络模型,依据Degree值筛选前 5 作为核心活性成分;通过 STRING 数据库搭建交集靶点的蛋白质-蛋白质相互作用(PPI)网络;将PPI网络导入Cytoscape 3.8.0,并使用CytoHubba插件对PPI网络进行分析,以Degree值前10的靶点为核心靶点;通过R语言插件对核心靶点进行GO分析与KEGG分析.利用"AutoDock Vina1.1.2"软件对核心靶点与核心活性成分进行分子对接.结果 筛选出通络解毒汤化学成分 127 个,靶点 442 个,AS疾病靶点 2 475 个,交集靶点 128 个.GO分析结果显示通络解毒汤治疗AS的生物过程主要涉及炎症反应,细胞凋亡与代谢、调节免疫等;KEGG分析结果显示主要通路为AGE-RAGE信号通路、IL-17 信号通路、HIF-1 信号通路等.分子对接进一步证实了核心靶点与核心活性成分的相互作用,其中TNF-α、MAPK1、MAPK3 与核心成分槲皮素、木犀草素、柚皮素、山柰酚等具有较强结合能力.结论 本研究预测吴门医派通络解毒汤可能是通过槲皮素、木犀草素、柚皮素、山柰酚等有效成分作用于TNF、CTNNB1、ESR1、MAPK3、MAPK1 等靶点调节多条信号通路,最终发挥其抗炎及调节免疫功能等作用.为后续进一步研究通络解毒汤治疗AS的分子机制提供依据及思路.
Based on compound network pharmacology and molecular docking technology,explore the mechanism of Wumen Tongluo Jiedu Decoction in the treatment of ankylosing spondylitis
Objective To explore the mechanism of Wumen Tongluo Jiedu Decoction in treating ankylosing spondylitis(AS)by network pharmacology and molecular docking technology.Methods TCMSP,TCMID and BATMAN-TCM databases were used to screen the related active ingredient and targets in Tongluo Jiedu Decoction;Collect AS related targets using GeneCards,Drugbank,and TTD databases;Obtain the intersection target of Tongluo Jiedu Decoction and AS using Venny2.1.0 software;construct network model of active ingredient-intersection targets,the top 5 were selected as the core active ingredients through the Degree value;Build a protein-protein interaction(PPI)network of intersecting targets through the STRING database;Import the PPI network into Cytoscape 3.8.0 and use the CytoHubba plugin to analyze the PPI network,with the top 10 targets with Degree values as the core targets;Perform GO analysis and KEGG analysis on core targets through the R language plugin.Use"AutoDock Vina1.1.2"software to perform Molecular Docking between core target and core active ingredient.Results 127 chemical components of Tongluo Jiedu Decoction were screened,with 442 targets,2 475 AS disease targets,and 128 intersection targets.The GO analysis results showed that the biological process of treating AS with Tongluo Jiedu Decoction mainly involves inflammatory reactions,cell apoptosis and metabolism,and immune regulation;the KEGG analysis results showed that the main pathways were AGE-RAGE signaling pathway,IL-17 signaling pathway,HIF-1 signaling pathway,etc.Molecular Docking further confirmed the interaction between the core target and the core active component,in which TNF-α、MAPK1 and MAPK3 have strong binding capacity with the core components Quercetin,Luteolin,Naringenin,Kaempferol,etc.Conclusion This study predicts that Wumen Tongluo Jiedu Decoction may regulate multiple signal pathways through the action of active ingredient such as Quercetin,Luteolin,Naringenin,Kaempferol,etc.on TNF,CTNNB1,ESR1,MAPK3,MAPK1 and other targets,and finally exert its anti-inflammatory and immune regulating functions.This study can provide basis and ideas for further research on the molecular mechanism of Tongluo Jiedu Decoction in treating AS in the future.
