小青龙汤变方在COPD大鼠气道炎症反应中的作用机制探讨

目的 研究慢性阻塞性肺疾病(COPD)大鼠模型中白细胞介素-6、肿瘤坏死因子-ɑ、干扰素-γ在大鼠血清和肺泡灌洗液中含量变化,探索我国传统经典方剂小青龙汤变方在COPD气道炎症治疗中的作用机制.方法 成年雄性SD大鼠 60 只,分为对照组 20 只,模型组 20 只,干预组 20 只,采用反复熏香烟+气管内注入脂多糖法建立COPD大鼠动物模型,其中干预组予灌胃小青龙汤变方.酶联免疫吸附法(ELISA法)检测大鼠血清、肺泡灌洗液中IL-6、TNF-ɑ和IFN-γ的含量.结果 在模型组中,血清和肺泡灌洗液中TNF-α和IL-6 较干预组均显著升高,IFN-γ表达下降.结论 COPD大鼠模型中,小青龙汤变方可能影响气道炎症因子水平,从而一定程度改善气道炎症.
Exploring the mechanism of the effect of Xiaoqinglong Decoction Modified Prescription on airway inflammatory response in COPD rats
Objective To study interleukin-6,tumor necrosis factor-ɑ and Interferon-γin rat model of chronic obstructive pulmonary disease(COPD),in order to explore the mechanism of Xiaoqinglong Decoction Modified Prescription in the treatment of COPD airway inflammation,the content changes in rat serum and alveolar lavage fluid were observed.Methods Sixty adult male SD rats were divided into the control group(20 rats),the model group(20 rats)and the intervention group(20 rats).The animal model of COPD rats was established by repeated cigarette smoking and intratracheal injection of lipopolysaccharide.The intervention group was fed with Xiaoqinglong Decoction Modified Prescription.Detection of IL-6,TNF-ɑ and IFN-γ in rat serum and alveolar lavage fluid by enzyme-linked immunosorbent assay content was measured by ELISA.Results In the model group,TNF-α in serum and alveolar lavage fluid and IL-6 significantly higher than the intervention group,IFN-γexpression decreased.Conclusion In the COPD rat model,the Xiaoqinglong Decoction Modified Prescription may affect the level of airway inflammatory factors,thus improve airway inflammation to a certain extent.
