基于网络药理学探讨四逆散对抑郁症、功能性消化不良和乳腺增生的"异病同治"作用机制研究

目的 运用网络药理学方法,对四逆散"异病同治"抑郁症、功能性消化不良和乳腺增生的物质基础及作用机制进行探讨研究。方法 通过TCMSP数据库获取四逆散可能的活性成分,利用Swiss Target Prediction数据库预测活性成分的潜在靶点;在Genecard、Disgenet、OMIM等数据库检索抑郁症、功能性消化不良和乳腺增生的相关靶点;通过STRING网站了解蛋白质互作关系并运用Cytoscape 3。8。0 软件进行可视化;利用DAVID数据库进行基因本体论(GO)分析和京都基因与基因组百科全书(KEGG)分析。结果 预测得到四逆散"异病同治"的活性成分 125 个和共同作用靶点 246 个,富集分析得到 569 个GO功能和 157 条信号通路(P<0。01,FDR<0。01),分析其作用机制可能与癌症、PI3K-Akt、内分泌耐药、HIF-1 等信号通路有关。结论 四逆散具有多成分、多靶点、多通路治疗抑郁症、功能性消化不良和乳腺增生的作用特点,为四逆散"异病同治"科学内涵的诠释提供新的研究思路与方法。
Mechanism of "treating different diseases with the same method" of Sini Powder in the treatment of depression,functional dyspepsia and breast hyperplasia based on network pharmacology
Objective The network pharmacology method was used to study the material basis and action mechanism of"treating different diseases with same method"of Sini Powder for depression,functional dyspepsia and breast hyperplasia.Methods The Sini Powder possible active components were obtained through the TCMSP database and potential targets of the active components were predicted using the Swiss Target Prediction database.The relevant targets of depression,functional dyspepsia and breast hyperplasia were searched in Genecard,Disgenet and OMIM databases.The protein interactions were understood through the STRING website and visualized using Cytoscape 3.8.0 software.Gene Ontology(GO)analysis and Kyoto Genomics and Genomics Encyclopedia(KEGG)analysis were performed using the DAVID database.Results It was predicted to obtain 125 active components and 246 common targets of"treating different diseases with same method"of Sini Powder,569 GO functions and 157 signaling pathways were obtained by enrichment analysis(P<0.01,FDR<0.01).The mechanism of action may be related to cancer,PI3K-Akt,endocrine resistance,and other signaling HIF-1 pathways.Conclusion The effect and characteristics of Sini Powder in depression,functional dyspepsia and breast hyperplasia treatment were in multi-ingredient,multi-target and multi-pathway,which would provide new research ideas and methods for the interpretation of the scientific connotation of Sini Powder on"treating different diseases with the same method".
