目的 建立HPLC法测定蜈蚣配方颗粒中 3,8-二羟基喹啉的含量。方法 采用Kromasil 100-5 C18 色谱柱(250 mm×4。6 mm,5 μm),以 10 mmol/L的磷酸二氢钾溶液-甲醇(68∶32)为流动相,流速为 1。0 ml/min,柱温为 30℃,检测波长为 252 nm。结果 HPLC色谱图中阴性对照无干扰;3,8-二羟基喹啉在 0。01163~1。395 mg/ml(r= 0。9997)范围内呈良好的线性关系,平均加样回收率为 99。4%(RSD=2。7%)。结论 该方法准确、稳定、可行,本文建立的方法经方法学验证可用于评价蜈蚣配方颗粒的质量。
Simultaneous determination of quinoline-3,8-diol in Centipede Formula Granules by HPLC
Objective To establish a HPLC method for simultaneous determination of quinoline-3,8-diol in Centipede Formula Granules.Methods The chromatographic separation was achieved on a Kromasil 100-5 C18(column 250 mm×4.6 mm,5 μm)and the column temperature was at 30℃.The mobile phase consisted of a water solution containing 10 mmol/L monopotassium phosphate and methyl alcohol with isoelution(68∶32)at a flow rate of 1.0 ml/min,and the detection wavelength was set at 252 nm.Results HPLC chromatograms revealed no interference due to negative reference.Quinoline-3,8-diol showed good linear relationships within the ranges of 0.01163~1.395 mg/ml(r= 0.9997),whose average recoveries was 99.4%(RSD=2.7%),respectively.Conclusion This accurate,stable and feasible method can be used for the quality control of Centipede Formula Granules according to the methodology validation.
