目的 建立高效液相色谱法同时测定不同生长年限九节茶不同部位中的 8 种成分含量变化。方法 色谱柱为Phenomenex Luna(C18)(250 mm×4。6 mm,5 μm),以 0。2%磷酸(A)-乙腈(B)为流动相,梯度洗脱(0~12 min,2%~11%B;12~25 min,11%~18%B;25~40 min,18%~40%B),流速 1。0 ml/min,检测波长为 210 nm和 325 nm,柱温 30℃,进样量 10 μl。结果 富马酸、新绿原酸、绿原酸、隐绿原酸、咖啡酸、异嗪皮啶、落新妇苷、迷迭香酸 8 种成分分别在 7。61~304。40 μg/ml(R2 = 0。999 7)、3。52~140。60 μg/ml(R2 = 1。000 0)、4。99~199。60 μg/ml(R2 = 1。000 0)、3。89~155。40 μg/ml(R2 = 1。000 0)、0。56~22。06 μg/ml(R2 = 0。999 9)、0。57~22。62 μg/ml(R2 = 1。000 0)、26。75~1 070。00 μg/ml(R2 = 1。000 0)、12。49~499。40 μg/ml(R2 = 0。999 9)范围内线性关系良好。不同添加水平加标回收率为 95。05%~99。26%,其相对标准偏差在 1。15%~1。93%之间。结论 九节茶在茎中富马酸、绿原酸、咖啡酸、落新妇苷、迷迭香酸的含量 1 年生>2 年生>3 年生;新绿原酸、隐绿原酸的含量 1 年生<2 年生<3 年生;异嗪皮啶的含量 1 年生>3 年生>2 年生。在叶中富马酸、新绿原酸、隐绿原酸、咖啡酸、异嗪皮啶、落新妇苷、迷迭香酸的含量 1 年生>2 年生>3 年生;绿原酸的含量 1 年生>3 年生>2 年生。
Comparison of eight components of Sarcandra glabru in different growth years and different parts
Objective To establish a HPLC method for simultaneous determination of eight components in different growth years and different parts of Sarcandra glabru.Methods The Phenomenex Luna C18(250 mm×4.6 mm,5μm)column was used,the mobile phase was 0.2%phosphoate acid(A)and acetonitrile(B),gradient elution(0~12 min,2%~11%B;12~25 min,11%~18%B;25~40 min,18%~40%B)was performed;the flow rate was 1.0 ml/min;the detection wavelength was 210 nm and 325 nm,the column temperature was 30℃,the injection volume was 10μl.Results Within the following ranges,the linear relationship of eight components were good respectively:7.61~304.40 μg/ml for fumaric acid(R2 = 0.999 7),3.52~140.60 μg/ml for neochlorogenic acid(R2 = 1.000 0),4.99~199.60 μg/ml for chlorogenic acid(R2 = 1.000 0),3.89~155.40 μg/ml for cryptochlorogenic acid(R2 = 1.000 0),0.56~22.06 μg/ml for caffeic acid(R2 = 0.999 9),0.57~22.62 μg/ml for isofraxidin(R2 = 1.000 0),26.75~1 070.00 μg/ml for astilbin(R2 = 1.000 0),12.49~499.40 μg/ml(R2 = 0.999 9)for rosmarinic acid(R2 = 1.000 0),the recovery rate of different addition levels was 95.05%~99.26%,and its relative standard deviation was between 1.15%and 1.93%.Conclusion In the stem,the content of fumaric acid,chlorogenic acid,caffeic acid,astilbin,rosmarinic acid were 1 year>2 years>3 years;the content of neochlorogenic acid,cryptochlorogenic acid were 1 year<2 years<3 years;the content of isofraxidin was 1 year>3 years>2 years.In the leaves,the content of fumaric acid,neochlorogenic acid,cryptochlorogenic acid,caffeic acid,isofraxidin,astilbin,rosmarinic acid were 1 year>2 year>3 years;the content of chlorogenic acid was 1 year>3 years>2 years.
