基于指纹图谱和多指标成分含量测定的陈皮、青皮质量评价

目的 建立陈皮、青皮指纹图谱及多指标成分含量测定方法,结合指纹图谱相似度评价、聚类分析(Cluster analysis,CA)和偏最小二乘判别分析(Partial Least Squares Discriminant Analysis,PLS-DA),对多批次不同产地陈皮、青皮进行质量评价。方法 采用Agilent ZORBAX SB C18 色谱柱(250 mm×4。6 mm,5 μm),以乙腈-0。1%磷酸水为流动相进行梯度洗脱,流速 1。0 ml/min,柱温 30℃,检测波长330 nm。对不同产地 10 批陈皮、24 批青皮建立指纹图谱,确定共有峰并进行相似度分析,同时测定芸香柚皮苷、橙皮苷、香蜂草苷、川陈皮素和橘皮素的含量。结果 10 批陈皮、24 批青皮的HPLC指纹图谱的相似度均在 0。9 以上,陈皮标定 11 个共有峰,青皮标定 13 个共有峰,共指认其中 7 个峰,分别为芸香柚皮苷、橙皮苷、新橙皮苷、香蜂草苷、川陈皮素、橘皮素、5-去甲川陈皮素。通过CA和PLS-DA可将 10批陈皮和 24 批青皮(12 批四花青皮和 12 批个青皮)划分为 2 类,且通过PLS-DA项下的VIP分析在陈皮、青皮共有成分(芸香柚皮苷、橙皮苷、香蜂草苷、川陈皮素、橘皮素含量分别为 4。6~10。82、44。77~131。31、1。31~4。34、0。33~5。72、0。08~3。54 mg/g)中,筛选出橙皮苷、香蜂草苷、橘皮素为陈皮和青皮共有的质量差异的标志性成分。结论 本研究建立的陈皮、青皮质量评价方法简便、灵敏、准确、重复性好,可为陈皮、青皮的质量评价提供依据。
Quality evaluation of Citri Reticulate Pericarpium and Citri Reticulate Pericarpium Viride based on fingerprint and multi index component content determination
Objective The fingerprint spectra and multi index component content determination of Citri Reticulate Pericarpium(CRP)and Citri Reticulate Pericarpium Viride(CRPV)were established,combined with fingerprint similarity evaluation,cluster analysis(CA),and partial least squares discriminant analysis(PLS-DA)to evaluate the quality of multiple batches of CRP and CRPV from different origins.Methods Using Agilent ZORBAX SB C18 chromatography column(250 mm×4.6 mm,5 μm)acetonitrile 0.1%phosphoric acid water as the mobile phase for gradient elution,with a flow rate of 1.0 ml/min,column temperature of 30℃,and detection wavelength of 330 nm.Establish fingerprint spectra for 10 batches of CPR and 24 batches of CRPV from different regions,identify common peaks,and perform similarity analysis.Results Simultaneously determine the contents of naringin,hesperidin,coumarin,quercetin and hesperidin.The similarity of HPLC fingerprint spectra for 10 batches of CRP and 24 batches of CRPV was above 0.9.11 common peaks were identified for CRP and 13 common peaks were identified for CRPV.A total of 7 peaks were identified,including narirutin,hesperidin,neohesperidin,neoponcrin,nobiletin,tangeritin and 5-demethynobiletin.10 batches of CRP and 24 batches of CRPV[12 batches of Fructus Citri Immaturus(FCI)and 12 batches of Pericarpium Citri Reticulatae Viride(PCRV)]can be divided into 2 categories through CA and PLS-DA.Through VIP analysis under PLS-DA,the common components of CRP and CRPV(narirutin,hesperidin,neoponcrin,nobiletin,tangeritin content are 4.6~10.82,44.77~131.31,1.31~4.34,0.33~5.72,0.08~3.54 mg/g,respec-tively)were screened,and hesperidin,neoponcrin and tangeritin were be regarded as chemotaxonomic markers of the common quality differences between CRP and CRPV.Conclusion The quality evaluation method for CRP and CRPV established in this study is simple,sensitive,accurate,and has good repeatability,which can provide a basis for the quality evaluation of CRP and CRPV.
