Study on the feasibility analysis of different bovine serum replacing human serum as blank substrate for determination of tacrolimus drug concentration by HPLC-MS/MS method
Objective Using tacrolimus as the model drug,the feasibility analysis of different bovine serum replacing human serum as blank substrate was investigated in the determination of drug concentration by HPLC-MS/MS.Methods Under the same treatment method,human serum and different bovine serum were used as blank matrix to prepare standard curves,and the concentration of 101 tacrolimus serum samples was com-pared under different standard curves.Results The relative deviation between the three types of bovine serum(calf serum,newborn bovine serum,fetal bovine serum)and human serum was greater than TEa(12.5%),and they could not replace human serum as blank substrate.Conclusion When tacrolimus was used as the model drug,none of the three bovine serum could replace human serum as the blank substrate in the determination of drug concentration by HPLC-MS/MS.
