摘要

目的 建立同时测定清热祛瘀液中丹皮酚、木香烃内酯和去氢木香烃内酯的反相高效液相色谱法.方法 采用CAPCELL PAK C18柱(250 mm×4.6 mm,5 μm),流动相为甲醇-水(70:30),流速1.0 ml/min,检测波长:215 nm,柱温为30℃,进样量为 10μl.结果 丹皮酚在浓度范围0.754 1~60.329 1μg/ml(r=0.999 6)、木香烃内酯在浓度范围1.156 3~92.507 4 μg/ml(r=0.999 9)、去氢木香烃内酯在浓度范围1.381 8~110.544 5 μg/ml(r=0.999 8),线性良好,三种物质检出限(LOD)为0.01~0.04μg/ml、定量限(LOQ)为0.03~0.13μg/ml;三种物质平均回收率为97.0%~103.5%,回收率RSD为1.3%~1.8%(n=6).结论 该方法操作简便,重复性好,准确度高,可用于清热祛瘀液的含量测定及质量控制.

Abstract

Objective A reverse-phase high-performance liquid chromatography method was established for the simultaneous determination of paeonol,coxylanolide and dehydrocoxylanolide in Qingre Quyu Liquids.Methods CAPCELL PAK C18 column(250 mm×4.6 mm,5 μm)were selected,mobile phase was methanol-water(70:30),flow rate was 1.0 ml/min,detection wavelength:215 nm,column temperature was 30℃,and sample intake was 10 μl.Results Paeonol,coxylanolide and dehydrocoxylanolide showed a good linear relationship in the range of 0.754 1~60.329 1μg/ml(r=0.999 6),1.156 3~92.507 4 μg/ml(r=0.999 9),1.381 8~110.544 5 μg/ml(r=0.999 8),the decection limit for the three substances was 0.01~0.04μg/ml,LOQ was 0.03~0.13μg/ml;the average recovery rate of the three substances were 97.0%~103.5%,the RSD were 1.3%~1.8%(n=6).Conclusion This method is easy to operate,with good repeatability and high accuracy,can be used for the content determination and quality control of Qingre Quyu Liquids.
