Study on the mechanism of berberine against novel coronavirus infection based on network pharmacology
Objective To preliminary predict the targets and signaling pathways of berberine against novel coronavirus infection by network pharmacology,and to preliminarily explore the mechanism of berberine.Methods The corresponding target sites of the drug were obtained from SwissTargetPrediction and PubChem databases.GeneCards and DisGenNET databases were used to collect disease-related targets.Venny website online tool was used to screen the intersection targets of the drug and disease,and the STRING database and Cytoscape 3.9.1 software were used to construct the"drug-target-disease"and PPI network diagram.GO and KEGG enrichment analysis of intersection targets were performed using DAVID database.Results A total of 50 intersection targets of berberine and novel coronavirus infection were obtained.The core PPI network was obtained by topological analysis,which contained 17 nodes.GO functional analysis showed that the biological processes of berberine in the treatment of novel coronavirus infection included:signal transduction,protein phosphorylation,and positive regulation of protein entry into the nucleus.Cellular components include:cytoplasm,plasma membrane,nuclear plasma.Molecular functions include:serine/threonine/tyrosine kinase activity,ATP binding,insulin receptor,etc.A total of 95 enriched pathways were obtained by KEGG pathway analysis,and the main signaling pathways related to novel coronavirus infection included VEGF signaling pathway,chemokine signaling pathway,Rap1 signaling pathway and PI3K-Akt signaling pathway.Conclusion Berberine has anti-novel coronavirus infection effect,which may exert its therapeutic effects by regulating VEGF signaling pathway,chemokine signaling pathway,Rap 1 signaling pathway and PI3K-Akt signaling pathway,thus regulating the immune response and inflammatory activation process.
