Application of modern analytical techniques in structural characterization of tetanus toxoid
Objective To characterize the structure of tetanus toxoid(TT)using modern analytical techniques.Methods Circular dichroism(CD)was used to investigate the secondary structure of TT,differential scanning calorimetry(DSC)was used to investigate the denaturation temperature of TT,then the determination results of different batches of samples were compared.Results 9 batches of TT α-helix ratio is 25.4%~26.2%.The anti-parallel is 12.3%~14.9%.The parallel is 6.4%~7.0%.The β-turn is 21.7%~24.0%,the random coil is 31.8%~33.9%,and the melting temperature is 89.19℃~90.47℃.The repeatability and inter batch consistency of the results are good.Conclusion Circular dichroism and differential scanning calorimetry can be used for structural characterization of tetanus toxoids,providing technical support for the stability and consistency of such products in the production process.
