Analysis on the Mechanism of Xiaochengqi Decoction in the Treatment of Horse Intestinal Obstruction Based on Network Pharmacology
Intestinal obstruction is one of the most common diseases in horses,and Xiaochengqi decoction is a classic formula for the treatment of intestinal obstruction.The aim of this study was to elucidate the mechanism of action of Xiaochengqi decoction for the treatment of equine intestinal obstruction by network pharmacological analysis.Firstly,all the chemical components and targets of Xiaochengqi decoction were searched in the systematic pharmacology data-base of traditional Chinese medicine,and then the compound-target network,protein-protein interaction(PPI)net-work,and target-pathway network were constructed by using STRING,DAVID,NCBI database,and Cytoscape soft-ware,to screen the active compounds and targets of Xiaochengqi decoction.The network diagram of Xiaochengqi de-coction was constructed,and the GO and KEGG pathways of Xiaochengqi decoction in the treatment of equine intesti-nal obstruction were studied and analyzed.The results were as follows:16 effective compounds and 32 corresponding targets were screened by OB,DL and Caco-2;the PPI network map of Xiaochengqi decoction acting on equine intesti-nal obstruction contained 45 targets,and the main key targets were TP53,ESR1,NCOA1,NCOA2,IGF1,MAPK14,etc,among which the targets directly related to equine intestinal obstruction were mainly TP53,ESR1,TGFB1,MMP9,HSP90AB1;there were 24 gene ontology(GO)entries and 7 KEGG pathways.The present study suggests that Xiao-chengqi decoction may regulate the expression of proteins such as TP53,ESR1,NCOA1,NCOA2,IGF1,and MAPK14 through zeylanaxanthin,cassia lactone,carotenoid qt,β-sitosterol,eudesmol,and isohespergone.It also treats equine intestinal obstruction through the tumour pathway,MAPK signalling pathway,tumour proteoglycan,and tumour necrosis factor signalling pathway.The mechanism of action needs further biological validation.
