Analysis on Expression Level of Ten Candidate Genes in Skin Tissue of Bashibai Sheep with Different Coat Color
In order to explore the relationship between ten candidate genes(TCF25,TYR,TYRP1,ASIP,KIT,MITF,EDNRB,SLC7A11,MC1R and DCT)that may affect animal coat color and Bashibai sheep coat color,qRT-PCR was used to study the mRNA expression of coat color related genes in the skin of 30 Bashibai sheep at the same year of age with different coat colors(10 black,10 white and 10 brown coat).All data were analyzed by SPSS 23.0 for one-way ANOVA.The results showed that the expression of TCF25,ASIP and EDNRB genes in skin of white coat Bashibai sheep was significantly higher than that in skin of black and brown coat(P<0.05),and the expression of TYR,TYRP1,DCT and MC1R genes in skin of black coat was significantly higher than that in skin of white and brown coat(P<0.05).The expression of MITF gene in skin of brown hair was significantly higher than that in skin of white and black(P<0.05),but there was no significant difference in KIT and SLC7A11 genes in different coat color skin(P>0.05).In summary,TCF25 and ASIP genes mainly related the white coat color traits of Bashbai sheep,and TYR,TYRP1,DCT and MC1R mainly related the black wool color traits of Bashbai sheep,while the specific mechanisms of MITF and EDNRB genes involved in regulating the brown and white coat color traits of Bashbai sheep need to be further explored.
