Antibacterial Activity of Pleuromutilin Derivative PCI Against 48 Clinical Isolates of Bovine Methicillin-resistant Staphylococcus Aureus
In order to investigate in vitro and in vivo antibacterial activity of the truncated pleuromutilin derivative PCI against 48 clinical strains of methicillin-resistant Staphylococcus aureus in bovine mastitis,the minimal inhibitory con-centration(MIC)of PCI against Staphylococcus aureus,methicillin-resistant Staphylococcus aureus(MRSA)standard strains,and 48 clinical strains of methicillin-resistant Staphylococcus aureus in bovine mastitis were determined by micro-broth dilution method,and the thigh infection experiment in mice was carried out.The results showed that the MIC of PCI against S.aureus and MRSA was 0.007 8µg/mL,and the MIC of PCI against bovine MRSA clinical strains was 0.007 8-0.125µg/mL.The results of thigh infection in mice showed that PCI reduced the degeneration and necro-sis of muscle tissue,and the amount of bacteria in thigh tissue was significantly lower than that in the tiamulin fumarate group(P<0.05),which was significantly lower than that in the model group(P<0.000 1).In summary,PCI has excellent antibacterial activity against 48 clinical isolates of methicillin-resistant Staphylococcus aureus in bovine mastitis,which can alleviate thigh muscle infections caused by MRSA.
