燃煤型氟斑牙动物模型复制方法的改进

摘要
目的 建立高仿真燃煤型大鼠氟斑牙动物模型.方法 以高氟煤拌粘土为燃料复制疫区空气环境,病区煤烘玉米配制饲料,30只SD大鼠按雌雄各半体重均等随机分为5组,即高氟空气饲养室内给予食物氟40 mg/kg (H)、25 mg/kg(M)、10 mg/kg(L)、2.1 mg/kg(A)组,阴性对照(C)组,观测大鼠体重、氟斑牙发生情况、尿氟及血氟含量、下切牙牙冠长度及釉质表面的超微结构.结果 生火期空气氟浓度为(28.5±5.3) μg/m3、闭火期为(4.1±1.2) μg/m3.高氟空气环境下各组雌、雄大鼠体重增长均较C组缓慢(P< 0.05).H、M组大鼠下切牙釉质出现白色条纹和白垩色斑等变化,尿氟、血氟含量较C组升高(P<0.05).H、M组下切牙冠长度增长较C组慢(P<0.05).釉质表面白垩色区域矿化不全、釉柱间隙增宽.结论 高氟空气环境对大鼠的生长发育可能会产生一定影响,通过模拟高氟空气环境并饲予煤烘玉米成功复制了高仿真的燃煤型氟斑牙动物模型.
Abstract
Objective To establish high-quality imitation of model replication of animal dental fluorosis caused by coal burning. Methods High fluoride air model was established on the basis of living habit of epidemic area by coal burning with clay riches of fluoride. Feeding rats with corn dried by coal burning at different percentage. Thirty SD rats ( 15 males and 15 females) were randomly divided into 5 groups: in the high fluoride air room and feed food with fluorine at 40 mg/kg group ( H) , 25 mg/ kg group (M) , l0mg/kg group ( L) , 2. 1 mg/kg group ( A) and the control groupf C). Recorded dental fluorosis progresses and measured rats' weight, tooth length, concentrations of fluoride in urine and plasma. Observed the surface of enamel by scanning electron microscope ( SEM). Result The fluoride concentration of air was (28. 5 ±5.3) ug/m3 at the burning time, and (4. 1 ±1.2) ug/mat other time. Separately compared with the male and the female of the group C, the mean of rats weight descended in the experimental group ( P < 0. 05). A succession of white and pigmented bands was observed in the enamel of mandibular incisors of the group H and M. The concentrations of fluoride in urine and plasma ascended compared with group C ( P < 0. 05 ). It showed a slow growth in the length of crowns of mandibular in group Hand M compared with group C ( P <0. 05). Observed crystal structure normal but the gap between the enamel rods widen at the white surface of fluorosis incisors by SEM. Conclusion High fluoride air model that would have effect on the rat growth. On these conditions, more model replication of animal dental fluorosis caused by coal burning could be established.
