全自动尿碘仪在尿碘、水碘和盐碘质控考核的应用分析

摘要
目的 获得一种快捷高效、准确灵敏的测定尿碘、水碘、盐碘检测方法.方法 通过全自动碘分析仪法对尿碘、水碘、盐碘质控样和考核样进行检测,分析全自动碘分析仪法标准系列的线性范围、检出限、加标回收率、准确度、精密度,并对仪器法与手工法检测结果进行t检验比较.结果 尿碘方法在0~300 μg/L标准曲线范围内吸光度的对数与浓度成线性,相关系数r=-0.999 9,检出限为3.5μg/L,2个浓度尿碘标准物质的测定值均在规定的不确定度范围内,平均加标回收率在91.2%~99.2%之间,相对标准偏差(RSD)为0.21%~1.62%(n=6).水碘方法在含碘量0~100μg/L标准曲线范围内吸光度的对数与浓度呈线性,相关系数r=-0.999 9,检出限为1.2μg/L,2个浓度水碘标准物质的测定值均在规定的不确定度范围内,平均加标回收率在92.7%~100.4%之间,RSD为0.35%~2.59%(n=6).盐碘方法在含碘量0~3.0mg/L标准曲线范围内吸光度的对数与浓度呈线性,相关系数r=0.999 8,检出限为0.26mg/kg,2个浓度盐碘标准物质的测定值均在规定的不确定度范围内,平均加标回收率在98.5%~102.1%之间,RSD为0.76%~1.02%(n=6).全自动尿碘仪测定尿、水、盐盲样的结果与传统手工方法测定值经t检验比较,差异无统计学意义(P>0.05).结论 该方法代替传统的手工法,操作方便快捷,灵敏度、准确度、精密度均符合检测要求.
Abstract
Objective This paper aims to obtain a fast,convenient,accurate and sensitive determination method for the test of urinary iodine,water iodine and salt iodine.Methods Urine iodine,water iodine and salt iodine were detected by automatic iodine analyzer and traditional manual methods,according to the design of the automatic urinaryiodine analyzer,validation experiments of the method were carried out on the detection limit,linear range,standard addition recovery,accuracy and precision,the results of the two methods were compared by t-test.Results The logarithm of absorbance was linear with the concentration in the standard curve range of iodine content(0-300 μg/L),with the correlation coefficient(r)greater than-0.999 9.The detection limit was 3.5 μg/L,and the determination values of the two concentrations of urine iodine reference substances were within the specified uncertainty range.The average recoveries ranged from 91.2%to 99.2%.Relative standard deviations(RSD)ranged from 0.21%to 1.62%(n=6).The logarithm of absorbance of water iodine method was linear with the concentration in the standard curve range of iodine content(0-100 μg/L),with the correlation coefficient(r)greater than 0.999 9.The detection limit was 1.2 μg/L,and the measured values of two concentrations of water iodine reference substances were all within the specified uncertainty range.The average recovery was 92.7%-100.4%.RSD ranged from 0.35%to 2.59%(n=6).The logarithm of absorbance of salt iodine method was linear with the concentration in the standard curve range of iodine content(0-3.0 mg/L),with the correlation coefficient(r)greater than-0.999 8.The detection limit was 0.26 mg/kg,and the measured values of two concentrations of salt iodine reference substances were all within the specified uncertainty range.The average recoveries ranged from 98.5%to 102.1%.RSD ranged from 0.76%to 1.02%(n=6).There was no significant difference between the results of blind samples of urine,water and salt determined by automatic urinary iodine analyzer and those determined by traditional manual method by t-test(P>0.05).Conclusion This method replaces the traditional manual method,it is convenient and fast in operation,and its sensitivity,accuracy and precision meet the detection requirements.
