Preparation and Application of Hyperimmune Serum of Epidemic Encephalitis B
In order to satisfy the needs of identification test and exogenous virus test for encephalitis B live vaccines,specific serum of encephalitis B was prepared by repeated immunization in SPF chickens taking live and inactivated vaccines as immunogens.When the serum titer was up to requirement,the serum was collected using the method for heart blood collection,after being packed and freeze-dried,then purity test,plaque reduction neutralization titer test,specificity test,residual water test and uniformity test were conducted,and identification test and exogenous virus test for encephalitis B live vaccines were carried.The results showed that the prepared positive serum was sterile,the test results of bacteria,Mycoplasma and exogenous viruses were all negative,and failed to crossly react with foot-and-mouth disease virus(FMDV),porcine circovirus(PCV),pseudorabies virus(PRV),classical swine fever virus(CSFV),porcine reproductive and respiratory syndrome virus(PRRSV),porcine parvovirus(PPV)and bovine viral diarrhea virus(BVDV).The titer of plaque reduction neutralization test was>1∶1 000,and the residual moisture was<3%,without obvious difference in the titer test results of 10 samples in intra and inter groups(P<0.05).In conclusion,the prepared serum was with high titer,less residual moisture,good purity,specificity and uniformity,and could meet the need of test for encephalitis B.
