Bioinformatics Analysis on VP27 Protein of Type Ⅱ Goose Astrovirus
In order to investigate the structure and function of VP27 protein of Type Ⅱ goose astrovirus(GAstV-Ⅱ),its strain of ZYL02 with good immunogenicity was analyzed using bioinformatics software.VP27 protein's physicochemical properties were predicted and analyzed by Expasy-ProtParam,PSORT Ⅱ Prediction,ProtScale,SignalP 4.1,TMHMM 2.0 and SOPMA software,and its B-cell antigenic epitopes were predicted and analyzed using BepiPred-2.0,ABCpred,IEDB,ToxinPred2 and VaxiJen v2.0 software,then the dominant antigenic epitopes were selected;the VP27 protein was modeled using Phyre2 software,and the distribution of the dominant epitopes in the model was accurately predicted using PyMOL software.The results showed that VP27 protein was unstable and hydrophilic with subcellular located in the nucleus,no transmembrane structural domains or signal peptides;in the secondary and tertiary structures,the average proportion of random curls,extended chains,spirals and β-turns were 41.91%,29.05%,22.41%and 6.64%,respectively;26 B-cell antigenic epitopes were predicted,and 4 B-cell dominant antigenic epitopes were identified and located at 26-41 aa(FGIPQADSRSRYNANI),48-57 aa(RGRTSTSFTL),111-126 aa(TSTGGQITELRNRLNI)and 174-189 aa(PVLINWSVSDSQEQYN);and it was found that,by the localization of the tertiary structure,all the 4 dominant B-cell epitopes were located on the surface of the protein.In conclusion,multiple potential sites for inducing humoral immunity were available in VP27 protein.The physicochemical properties and structure of VP27 protein were predicted by bioinformatics analysis,contributing to further researches on the functions of GAstV-Ⅱ VP27 protein and the development of corresponding vaccines.
