An Effective Method for Collecting Nucleic Acid Aerosols
The purpose of this experiment was to determine an effective method to collect nucleic acid aerosols for monitoring nucleic acid aerosol contamination in laboratory.100 μL pBR322 plasmid DNA was evenly sprayed into a room with a volume of 23 m3 to simulate nucleic acid aerosol contamination.0(i.e.the day of spraying)and 5 days later,samples were collected from six sampling sites by the methods for control group and experimental group,and detected by real-time PCR.The samples in control group was sampled with a centrifuge tube filled with ultrapure water,and those in experimental group was sampled by gas sampling pump connecting filter tip.The results showed that,at 0 day,the Ct Mean of the experimental group from six sites was 22.732,lower than that of the control group that was 25.027;5 days later,Ct values were detected from only three sites in the control group,all of which were higher than those in corresponding sites in experimental group.In the routine monitoring of nucleic acid aerosols in laboratories of Brucella molecular biology,the method for experimental group was also with higher sensitivity.In conclusion,the method for experimental group was efficient for sampling,and suitable for monitoring nucleic acid aerosol contamination in laboratory.The monitoring of nucleic acid aerosol contamination in laboratory was technically supported by the study.
