Correlation Analysis on HI Assay and Commercial ELISA Kit for Antibodies Against Newcastle Disease Virus
In order to compare the correlation between Hemagglutination inhibition(HI)assay and a commercial ELISA kit for antibodies against Newcastle disease virus(NDV),and to explore determination criteria of NDV ELISA kit in compliance with China's immunization policy,267 clinical serum samples as well as standard negative or positive serum samples were detected using HI assay and ELISA kit with a foreign brand,respectively,to explore the correlation coefficient of the two methods,and the results by ELISA were analyzed using TG-ROC method taking HI assay as a gold standard to determine the optimal ELISA critical value for antibody detection of serum samples under the status of domestic immunological conditions.The results showed that the two methods had a good correlation with a correlation coefficient of 0.982 3;both the sensitivity and specificity of ELISA could reach 95.5%when the antibody potency≥8 400 was identified as"qualified immunization".In conclusion,the ELISA kit was significantly correlated with HI assay,particularly for detection of clinical serum samples on a large scale,but their critical value should be reduced in the context of immunization policy implemented in China.The correlation between HI assay and ELISA kit was analyzed in the study,which laid a foundation for detection of antibodies against NDV on a large scale using ELISA.
