Expression and correlation study of circRNA00809 and HES1 in the placental tissue of patients with hypertension in pregnancy
Objective To investigate the expression and correlation of circRNA00809 and hairy and enhancer of split 1 in the placental tissue of patients with hypertension in pregnancy.Methods A total of 25 healthy pregnant women and 25 patients with hypertension in preg-nancy admitted to the Obstetrics and Gynecology Department of the Special Medical Center of the Armed Police Force from January 2018 to July 2020 were selected as the normal control group and the hypertension in pregnancy group,respectively.General data of both groups were collected.The placental weight,neonatal birth weight,liver function,and renal function indicators were compared between the two groups.Placental tissues obtained after cesarean section were taken from both groups.RT-PCR was used to detect the expression levels of cir-cRNA00809 and HES1 mRNA,and the correlation between circRNA00809 and HES1 mRNA expression was analyzed.The expression levels of HES1 protein in the placental tissues of each group were determined by Western blotting.Results There were no significant differences in parity,age,pre-pregnancy body mass index gestational age at delivery,creatinine,blood urea nitrogen,platelets,and alanine aminotrans-ferase between the two groups(all P>0.05);however,the placental weight,neonatal birth weight,and 24-hour proteinuria were signifi-cantly lower in the hypertension in pregnancy group than in the normal control group(all P<0.05),while systolic pressure,diastolic pres-sure,and uric acid were significantly higher(P<0.05).The expression level of circRNA00809 in the placental tissue of the hypertension in pregnancy group(2.335±0.134)was significantly higher than that in the normal control group(0.831±0.072),with a statistically signifi-cant difference(P<0.05).The expression level of HES1 mRNA in the placental tissue of the hypertension in pregnancy group(0.492±0.208)was significantly lower than that in the normal control group(1.781±0.310),with a statistically significant difference(P<0.05).The correlation analysis showed a negative correlation between the expression levels of circRNA00809 and HES1 mRNA in the placental tis-sue(r=-0.477,P<0.05).The WB results showed that the expression level of HES1 protein in the placental tissue of the hypertension in pregnancy group was significantly lower than that in the normal control group,with a statistically significant difference(P<0.05).Conclusion circRNA00809 is highly expressed and HES1 is lowly expressed in the placenta of patients with hypertension in pregnancy.circRNA00809 may participate in the occurrence of hypertension in pregnancy through negative regulation of HES1 expression.
