摘要

目的 通过比较瑞博祥、新海生物、位点、博坤、微略五种国产手工磁珠试剂盒在DNA回收效率、提取质量方面的差异,为实践中如何选择试剂盒提供参考.方法 用五种国产磁珠试剂盒提取0.1 ng、0.2 ng、0.4 ng、0.6 ng、0.8 ng、1 ng 007 标准品(0.1 ng/μL),对提取的DNA进行定量和STR检测分析,比较五种试剂盒对标准品DNA的回收率和提取质量.结果 当007标准品的含量为0.1 ng、0.2 ng、0.4 ng时,五种试剂盒的回收率无显著性差异,但STR分型结果存在区别;当标准品含量为0.6 ng及以上时,五种试剂盒的DNA回收率开始出现差异,但STR分型结果差异不明显.结论 DNA含量>0.6 ng时,上述任意一种试剂盒提取DNA均能满足工作需要,但当DNA含量较少时,新海生物磁珠试剂盒进行提取纯化后STR分型结果最佳.

Abstract

Objective To compare the DNA extraction efficiency of five common domestic magnetic bead kits from Ruiboxiang,Xinhai Biological,Wei Dian,Bo Kun,and Wei Lue,and then provide a reference for how to choose these kits in practice.Methods Five domestic magnetic beads kits mentioned above were used to extract 007 standards(0.1 ng/μL)with the contents of 0.1 ng,0.2 ng,0.4 ng,0.6 ng,0.8 ng and 1 ng.Then the extracted DNA was quantified followed by STR analysis to compare the collecting efficiency and extraction quality of the five kits.Results When the DNA contents were 0.1 ng,0.2 ng and 0.4 ng,there were no significant differences in the recovery rate of the five kits,but the differences can be seen in STR typing results,while for 0.6 ng and above contents,the difference among the five kits begin to appear.Conclusion Any of the above kits can be used to extract DNA with the contents more than 0.6ng,but for few DNA contents,it is recommended to extracting and purifying by Xinhai biological kit.
