Viral genome characteristics and traceability analysis of a COVID-19 epidemic in Hebei Province
Objective To analyze the whole genome characteristics of SARS-CoV-2 in local COVID-19 epidemic,and to carry out traceability investigation based on epidemiology,so as to provide technical support for the accurate prevention and control of the epidemic.Methods The throat swab samples were collected from some confirmed cases of COVID-19 in a local epidemic in Hebei Province from October to November 2021,the whole genome of the viruses were sequenced by high-throughput sequencing technology to analyze the variation characteristics of viral nucleotide and amino acid sequences.Results A total of 68 high-quality virus genome sequences related to this epidemic were obtained,all of which were the evolutionary branch of Delta variant AY.126.After comparison,the first case sequence was highly homologous with the virus sequence of the Ejina Banner epidemic in Inner Mongolia during the same period,and the total number of nucleotide mutations in 68 sequences was 44-48 compared with Wuhan reference strain(NC_045512),the nucleotide mutation type was converted to primary by T.On S protein,10 amino acid mutations such as D614G,L452R,T478K,P681R and 2 amino acid deletions were found.Conclusion SARS-CoV-2 whole genome sequencing for early cases and key cases could quickly indicate the direction of epidemiological investigation,and tracked the characteristics of virus gene variation,which could provide strong technical support for epidemic prevention and control.
