Application of DNA barcoding technology for identification of imported murine animal at ports
Objective To explore the feasibility of DNA barcoding technique for identification of rat species,which is difficult to identify morphologically.Methods One murine animals imported from Jiangsu ports that was morphologically difficult to identify was collected.Mitochondrial cytochrome C oxidase Ⅰ(COⅠ)gene was amplified by PCR method,the target fragment was sequenced for sequence comparison analysis,and the genetic distance between species and within species was calculated.Results The sequence comparison showed that the imported rodent was Rattus rattus.Phylogenetic analysis showed that the intraspecific genetic distance was 7.7%,and the interspecific genetic distance was 11.6%~21.0%.The imported murine animal formed one independent evolutionary branch on the evolutionary tree.Conclusion DNA barcoding technology could accurately and effectively identify imported murine animals,especially help to improve the accuracy of identification of morphologically difficult murine animals.
