Genomic characterization of the first detected imported monkeypox virus at Zhuhai port
Objective To investigate the genomic characteristics and genetic variation of the first imported monkeypox virus(MPXV)at Zhuhai port,with the aim to provide technical support for the prevention and control of Mpox at ports.Methods The nucleic acid was extracted from oral swab sample of the first imported MPXV infected arriving passenger at Zhuhai port.The whole genome of the virus was amplified using multiplex PCR method provided by the MPXV NGS library construction auxiliary kit and sequenced by nanopore third-generation sequencer.The bioinformatics softwares were applied to analyze the genetic characteristics,subtype and mutations of the genome.Results The sequence length of the first imported MPXV at Zhuhai port was 197 177 nt,and the nucleotide homology with the reference sequence(NC_063383.1)was 95.7%.Compared with the reference genome(NC_063383.1),there were 36 amino acid mutations,involving multiple functional changes such as host immune regulation and virus surface structure.Phylogenetic analysis showed that the virus clustered with LC760460(Japan,2023),OR855744(South Korea,2023)and LC753968(Japan,2022)tightly,all of which belonged to the Ⅱb C.1 lineage.Conclusion The MPXV detected for the first time at Zhuhai port belonged to Ⅱb C.1 lineage and contained 36 amino acid mutations,calling for the continuous surveillance of MPXV and its genomic variation.
