Genetic diversity analysis of Haemaphysalis longgicornis in Yancheng based on mitochondrial gene COⅠ identification technology
Objective To determine the genetic diversity of Haemaphysalis longgicornis and phylogenetic relationships among different geographic populations,in order to provide a basis for the prevention and control of H.longgicornis and tick-borne diseases.Methods Morphological identification was conducted on ticks collected from Sheyang and Dongtai of Yancheng City from October 2022 to April 2023,including 6 kinds of habitats such as goji berry shrubs,saline alkali marsh areas,seawall grass,white thatch,residential areas in Tinghu,and riverbank grass in Dongtai.The molecular identification was performed through conventional PCR for amplifying the mitochondrial cytochrome C oxi-dase Ⅰ subunit(COⅠ)gene.Results The 38 collected ticks(25 adult ticks and 13 nymphs)in this study were i-dentified as H.longgicornis by morphology and molecular biology methods.The H.longicornis collected in Yancheng were distributed in two evolutionary branches,with the main evolutionary branch located in the"Compound"branch.Different geographical populations belonged to 5 shared haplotypes,with 25 being exclusive haplotypes.Among them,Yancheng was involved in a total of 3 shared haplotypes and 10 being exclusive haplotypes.In the network of hap-lotypes,H5 and H10 were found to be the primary haplotypes,they formed two radical centers,the representative se-quence haplotypes in Yancheng were rich and colorful.Conclusion There was no significant differentiation among different geographical populations in this study,and H.longicornis population in Yancheng might be undergoing the accumulation of expansion and mutation.
