Value of fluorescence quantitative PCR and auramine O staining in tuberculosis infection diagnosis
Objective To analyze the diagnostic value of quantitative fluorescence PCR and auramine O staining in tuberculosis infection,to improve the detection rate of Mycobacterium tuberculosis in tissue sections.Methods A to-tal of 100 archived wax blocks from patients suspected of tuberculosis infection,admitted to the 900th Hospital of Joint Logistics Support Force between June 2019 and December 2022,were selected as specimens.Both quantitative fluorescence PCR and auramine O staining were employed to detect Mycobacterium tuberculosis.The operational time of each method was compared,the results along with diagnostic efficacy for single tests versus combined testing were analyzed.Results The operational time for auramine O staining was(27.06±2.78)minutes,significantly shorter than that for quantitative fluorescence PCR at(56.56±3.62)minutes(P<0.05).The sensitivity of combined auramine O staining and quantitative fluorescence PCR was found to be 94.67%(71/75),specificity was 88.00%(22/25),and ac-curacy reached 93.00%(93/100),all surpassing those obtained by either method alone quantitative fluorescence PCR or auramine O staining alone.ROC curve analysis indicated that the predictive value for detecting Mycobacterium tuberculosis infection using combined testing had an area under the curve(AUC)of 0.900,which was higher than that observed with quantitative fluorescence PCR(AUC=0.750)or auramine O staining alone(AUC=0.722)(P<0.05).Conclusion The combination of quantitative fluorescence PCR and auramine O staining could serve as an auxiliary method conducive to early diagnosis of tuberculosis infection.
