不同修饰纳米磁珠对病原菌快速富集的效果评价

目的 为满足口岸快速检测要求,筛选对所有病原菌均具有良好吸附效果的纳米磁珠,对不同修饰纳米磁珠快速吸附性能进行测试。方法 测定11种不同化学修饰的纳米磁珠对大肠杆菌和金黄色葡萄球菌快速吸附能力,获得具有良好吸附能力的磁珠,利用扫描电镜、透射电镜和傅里叶红外光谱仪测定对其均一性、粒径、修饰方式等性能。结果 Dyna磁珠对大肠杆菌和金黄色葡萄球菌的富集效率分别为(78。01±0。57)%和(71。98±1。19)%,Si-PSA磁珠为(57。28±0。66)%和(89。10±2。40)%。Dyna磁珠粒径约500 nm,Si-PSA磁珠粒径约200 nm,粒径均一性较好。结论 筛选到对两种病原菌均具有良好吸附效果的纳米磁珠,可用于病原菌快速富集。
Evaluation of nanomagnetic beads with different modification on the rapid enrichment of pathogenic bacteria
Objective In order to meet the requirements of rapid detection at ports,nano magnetic beads with good adsorption effect on all pathogenic bacteria were screened,and the rapid adsorption performance of nano magnetic beads with different modifications was tested.Methods The rapid adsorption capacity of 11 chemically modified nano-magnetic beads for Escherichia coli and Staphylococcus aureus was determined,the magnetic beads with good adsorption function were obtained.The homogeneity,particle size and modification way of the nano magnetic were determined by scanning electron microscopy,transimission electron microscopy and Fourier transform infrared spectroscopy.Results The enrichment efficiency of Dyna magnetic beads for Escherichia coli and Staphylococcus aureus was(78.01±0.57)%and(71.98±1.19)%,respectively,and that of Si-PSA magnetic beads was(57.28±0.66)%and(89.10±2.40)%,respectively.The particle size of Dyna magnetic beads was about 500 nm,and the particle size of Si-PSA magnetic beads was about 200 nm.The particle size uniformity of both nano magnetic beads was good.Conclusion The nano magnetic beads with good adsorption effect on both pathogens were screened and could be used for rapid enrichment of pathogenic bacteria.
