利用DNA条形码测序技术鉴定高海拔地区口岸蚊虫种类

目的 利用DNA条形码技术对高海拔地区口岸蚊虫种类进行鉴定,为蚊虫分类及虫媒病毒病的防控提供科学依据。方法 2023年7-9月,在西宁曹家堡机场和格尔木机场的草坪、污水处理厂、宿舍等3个区域,采用紫外线诱蚊灯法和双层叠帐法捕蚊,利用形态学鉴定和DNA条形码测序鉴定蚊种。结果 共捕获蚊类110只,其中曹家堡机场口岸76只,格尔木机场口岸34只,形态学鉴定均为淡色库蚊。随机选择其中15只进行DNA条形码分子测序,序列比对证实14只为尖音库蚊,1只为致倦库蚊。结论 利用DNA条形码基因测序初步了解了高海拔地区口岸蚊种构成,更加精准地为口岸虫媒防控提供科学依据。
Identification of mosquito species collected at ports in high altitude by using DNA barcoding sequencing technology
Objective To identify the mosquitoes at high altitude ports by using DNA barcoding technology,and to provide scientific basis for mosquito classification and prevention and control of arboviral diseases.Methods From July to September 2023,mosquitoes were caught by UV light and double-layer tents at the lawn,sewage treatment plant and dormitory of Caojiapu Airport in Xining and Golmud Airport.The mosquitoes were identified by morpho-logical identification and DNA barcoding sequencing identification.Results A total of 110 mosquitoes were cap-tured,including 76 collected at Caojiapu Airport and 34 collected at Golmud Airport.All the species were identified as Culex pipiens pallens by morphological identification.Fifteen mosquitoes were randomly selected and DNA bar-coding molecular sequencing was performed.Sequencing blast showed that 14 were identified as Culex pipiens and 1 was Culex quinquefasciatus.Conclusion Using DNA barcoding sequencing molecular identification method,the pre-liminary exploration of mosquito species composition at ports in high-altitude was understood,provided more accurate scientific basis for medical vectors prevention and control at ports.
