富血小板纤维蛋白对大鼠骺板软骨细胞增殖和分化的影响

目的 探讨富血小板纤维蛋白(platelet-rich fibrin,PRF)对体外培养大鼠骺板软骨细胞增殖和分化的影响.方法 选取 2 周龄 SD 大鼠 4 只,获取原代骺板细胞后进行培养及鉴定.采集 8 周龄大鼠全血 10 ml,制备 PRF.实验分组:对照组在培养基中不加 PRF;实验组在培养基底部加入 PRF 膜.采用CCK-8、划痕实验、硫酸糖胺聚糖含量(glycosaminoglycan,GAG)检测评价在 PRF 作用下细胞的增殖、迁移、生物学功能活性.结果 PRF 作用后 48 h,CCK-8 检测显示实验组增殖率较对照组显著升高,差异有统计学意义(P<0.05).PRF 作用 24 h 和 48 h,划痕实验提示实验组伤口闭合度分别为(81.27±3.02)%、(97.60±0.89)%,较对照组(28.04±4.50)%、(45.68±2.44)%显著升高,对比差异有统计学意义(P<0.05).实验组 PRF 作用 24 h,与对照组相比 GAG 含量差异无统计学意义(P>0.05);PRF 作用 48 h,实验组的 GAG含量为(102.45±4.11)μg/ml,与对照组(56.62±4.61)μg/ml相比显著升高,差异有统计学意义(P<0.05).结论 PRF可以促进骺板软骨细胞增殖、分化,还可以促进 GAG的产生.
Effect of platelet-rich fibrin on the proliferation and differentiation of rat epiphyseal plate chondrocytes
Objective To study the effect of platelet-rich fibrin(PRF)on the proliferation and differentiation of rat epiphyseal plate chondrocytes cultured in vitro.Methods Four 2-week-old SD rats were selected,and primary epiphyseal plate cells were obtained and subjected for culture and identification.10 ml whole blood of 8-week-old rats was collected to prepare PRF.PRF was not added in the medium of the control group;PRF membrane was added at the bottom of the medium of the experimental group.CCK-8,scratch experiment and sulfate glycosamine polysaccharide content glycosaminoglycan(GAG)were used to evaluate the proliferation,migration and biological function activity of cells with PRF.Results After 48h of PRF treatment in the experimental group,the CCK-8 test showed a significantly higher proliferation rate compared with the control group(P<0.05).After 24 h and 48 h of PRF treatment,the scratch experiment indicated that the wound closure of experimental group were(81.27±3.02)%,(97.60±0.89)%,which were significantly higher than the control group[(28.04±4.50)%,(45.68±2.44)%](P<0.05).After 24 h of PRF treatment in the experimental group,there was no statistical difference in GAG content compared with the control group(P>0.05).After 48 h of PRF treatment in the experimental group,the GAG content(102.45±4.11)μg/ml was significantly increased than that of the control group(56.62±4.61)μg/ml(P<0.05).Conclusions PRF can promote the proliferation and differentiation of epiphyseal plate chondrocytes and GAG production.
