Research of the secondary metabolites from Arctic-derived fungus Penicillium sp.MYA5
Objective To investigate the secondary metabolites of fungi Penicillium sp.MYA5,isolated from the Arctic-Svalbard Archipelago.Methods The ethyl acetate extract of the fermented liquid extract was purified systematically after large scale fermentation,using normal phase silica gel column chromatography(HPLC),reversed phase ODS column chromatography and high performance liquid chromatography analysis methods.The structures of the compounds were identified on the basis of nuclear magnetic(NMR),mass spectrometry(MS)polarimetry,electronic circular dichroism(ECD)spectrum and the related literature.Results Seven compounds were obtained and identified as penicophenone C(1),communol G(2),1-(2,4-dihydroxy-5-methylphenyl)ethan-1-one(3),penicophenone A(4),conidiogenones C(5),cis-3,4-dihydro-3,4,8-trihydroxynaphthalen-1(2H)-one(6),(-)-(3S)-3-hydroxy-3-methyloxindole(7).Compound 1 was a new chroman derivative,an isomer of the known compound,(Z)-6-acetyl-3-(1,2-dihydroxypropylidene)-5-hydroxy-8-methylchroman-2-one,which was determined the absolute configurations for the first time.Compound 5 exhibited significant inhibitory on HL-60 and BEL-7402 with IC50 values of 0.44 and 1.16 μmol/L.
