摘要

目的 研究不同褐藻来源β-葡聚糖的结构及免疫活性差异,为β-葡聚糖药物的开发提供合适的藻源.方法 通过热水提取、乙醇沉淀和柱层析分离从6种褐藻中制备β-葡聚糖,并利用高效液相色谱(HPLC)、红外光谱(IR)、核磁共振波谱(1H NMR,13C NMR和1H 13C HSQC)对其结构进行表征.通过小鼠巨噬细胞RAW264.7吞噬中性红及NO释放实验评价其免疫调节活性.结果 本文制备了6种结构不同的β-葡聚糖,对其结构分析,证明其含有β-1,3和β-1,6连接,相对分子质量为2 290~5 490,海茸中β-葡聚糖的含量最高且具有最优的免疫活性,是β-葡聚糖药物开发的良好资源.

Abstract

Objective To investigate the structure and immunological activity of β-glucan from six kinds of brown algae,and provide the algae source for the development of marine carbohydrate-based drugs.Methodsβ-glucans were prepared from 6 kinds of brown algae by hot water extraction,ethanol precipitation and column chromatography.Their structures were characterized by high performance liquid chromatography(HPLC),infrared radiation(IR)and nuclear magnetic resonance spectroscopy(1H NMR,13C NMR and 1H 13C HSQC),and immune activities were evaluated by phagocytosis of neutral red and NO release amount of mouse macrophage RAW264.7.Results In this paper,six kinds of β-glucans with different structures were prepared,and their structures were analyzed.It was proved that they contained β-1,3 and β-1,6 linkages,and their relative molecular weights were 2 290~5 490,Durvillaea antarctica,which has the highest content of β-glucan and the best immune activity,is an ideal resource for β-glucan drug development.
