Objective To investigate the expression level and significance of microRNA-50-5p(miR-150-5p)and tumor necrosis factor receptor related factor 6(TRAF6)in serum of children with myasthenia gravis(MG).Methods From June 2021 to October 2022,60 MG children(observation group)and 60 healthy children(control group)were selected.The severity of MG was evaluated by quantitative myasthenia gravis score(QMG),and MG was classified by modified Osserman classification.The expression level of miR-150-5p in serum was detected by RT-PCR,and serum TRAF6,acetylcholine receptor(AchR)antibody,interleukin-4(IL-4)γ-Interferon(IFN-γ)The levels of plasma cell like dendritic cells(pDC)and regulatory T cells(Treg)were detected by flow cytometry.Spearman rank correlation analysis for correlation analysis.The diagnostic value of serum miR-150-5p and TRAF6 in MG was analyzed with the subject operating characteristic curve(ROC),and the area under the curve(AUC),sensitivity and specificity were calculated.Results Compared with the control group,in the observed group,miR-150-5p,TRAF6,AchR antibody,and IFN-γ levels were higher,while pDC,Treg,and IL-4 were lower(t=7.883,10.458,44.745,5.344,22.346,8.428,6.116,P<0.05).In the serum of MG children,the level of miR-150-5p was positively correlated with the level of TRAF6,AchR antibody and IFN-γ levels,respectively(r=0.576,0.456,0.470,P<0.05),but negatively associated with pDC,Treg,and IL-4 levels,respectively(r=-0.610,-0.454,-0.584,P<0.05).The TRAF6 levels were positively correlated with AchR antibody and IFN-γ levels,respectively(r=0.576,0.456,0.470,P<0.05),but negatively associated with pDC,Treg,and IL-4 levels,respectively(r=-0.432,-0.612,-0.512,P<0.05).The higher the MG subtyping,the higher the miR-150-5p and TRAF6 levels(r=0.664,0.589,P<0.05).Serum miR-150-5p and TRAF6 levels were positively correlated with the QMG scores,respectively(F=3.694,6.487,P<0.05).The combination of miR-150-5p with TRAF6 diagnosed MG with an AUC of 0.924,a sensitivity of 93.23%,and a specificity of 83.23%.Conclusion Serum miR-150-5p and TRAF6 expression levels were increased in children with MG,and both were associated with immune damage,and both contributed to the early diagnosis of MG.
