Isolation,Identification and Serologic Relativity of Two Strains of Goose-origin Reovirus
To understand the molecular characteristics and serological correlation of goose reovirus(GRV),goose embryo fi-broblast cells were inoculated with RT-PCR-positive liver tissue suspension of sick goose for virus isolation,and the pathogen's complete genome sequence was obtained by RT-PCR and sequencing.ST,LMH and Vero cells were infected with the isolates in order to investigate their growth characteristics in different cells.The cross-neutralization test was conducted to determine the se-rological association among waterfowl reoviruses.The results showed that two strains,named GD208 and GD225,were isolated from the liver tissue of sick goose,and the nucleotide homology between these two strains was low.The genetic distance between GD208 and the reference strain GRV-GD2020 was close,and the nucleotide homology was 99.1%to 99.6%.GD225 was genetically close to the novel duck reovirus(NDRV)such as XT18,DE150,QR strains reported since 2018,and the nucleotide homology was 95.3%to 98.9%.GD208 and GD225 proliferated best in LMH cells with TCID50 of 10-3.7/0.1 mL and 10-5.0/0.1 mL.In addition,GD208 could be neutralized by homologous serum and Muscovy duck reovirus(MDRV)positive serum,while GD225 could be neutralized by homologous serum,NDRV,MDRV and GD208 positive serum.In conclusion,two strains of GRV with different genotypes and serotypes were isolated in this study,and their molecular characteristics and antigenic correlation were different from other waterfowl reovirus,which could provide theoretical reference for the prevention and control of GRV infection.
