Effects of Different Antioxidants on Sperm Motility and Artificial Insemination Effect of Chicken Preserved in Vitro
In order to formulate a highly efficient diluent for the preservation of chicken semen,on the basis of the two for-mulas with better effects screened in the early stage,the three antioxidants of icariin,cysteine and chlorogenic acid were screened,and then artificial insemination experiments were performed with the selected diluent.The results showed that in the antioxidant screening experiments,formula A with cysteine added provided the best sperm preservation.At 48 h,formula A exhibited an aver-age motility of sperm surpassing 0.50,accompanied by significantly enhanced plasma membrane integrity rate and Total antioxi-dant capacity(T-AOC)concentration in comparison to the other formulas(P<0.05),while the sperm malformation rate in formula A was markedly lower than that in other formulas(P<0.05).The above results underscore the superiority of Formula A among the examined formulas,thus this formula was selected for artificial insemination experiments with diluted chicken semen.In the out-come of chicken farm insemination experiment,it was noted that at the onset of semen preservation(0 h),there were no significant differences in fertilization rate,hatching rate and chick survival rate between the three proportional dilution groups and the origi-nal sperm insemination group(P>0.05).At 8 h,the fertilization rate of eggs with 1∶3 ratio(83.50%)was significantly lower than that of 1∶1(93.60%)and 1∶2(92.94%)(P<0.05),while there was no significant difference in the hatch rate and chick surviv-al rate among the various dilution ratios(P>0.05).At 24 h,the fertilization rate,hatch rate and chick survival rate of eggs across all dilution ratios decreased significantly.The fertilization rate of eggs in the 1∶1 ratio group was significantly higher than that of the 1∶2 and 1∶3 ratio(P<0.05),the hatch rate and chick survival rate of the 1∶1 and 1∶2 ratio groups were significantly higher than that of the 1∶3 ratio(P<0.05),while there was no significant difference in the hatch rate and chick survival rate between the 1∶1 and 1∶2 ratio groups(P>0.05).The results indicated that the sperm dilution of formula A with cysteine had the best preservation effect,and the semen of chickens was preserved in vitro using self-developed dilution and semen in the dilution ratio of 1∶1 and 1∶2,and the sperm infusion at 0 h and 8 h could obtain better artificial insemination effect.
