Tissue Expression Profile,SNP Detection of ADSL Gene and Its Association Analysis with Meat Quality in Pigeon
In order to study the tissue expression profile of adenylosuccinate lyase(ADSL)gnen and its impact on meat quality traits,Experiment 1:16 tissues of 6 White King pigeons were used as materials for tissue expression profile analysis using Real Time Quantitative PCR.Experiment 2:140 individuals of White King pigeon were used as materials to screen for ADSL gene sequence SNPs using direct sequencing method.The pectoral muscles of these 140 individuals were collected for measurement of meat quality indicators such as shear strength,flesh color,pH value and electrical conductivity.The intramuscular fat content was measured using Soxhlet extraction method;Use gas chromatography-mass spectrometry to determine the content of fatty acids.The results showed that the ADSL gene exhibited a constitutive expression pattern,with the highest expression in the liver,chest muscle and leg muscle.There were 11 mutation sites in the sequence,among which T2305C,G2476A,C2664T SNPs were significantly or extremely correlated with polyunsaturated fatty acids,intramuscular fat and inosine monophosphate.The results indicated that the ADSL gene might be related to meat quality traits and these SNPs of ADSL genes could serve as molecular markers for meat quality traits,providing basic data for breeding meat pigeons.
