基于线粒体基因COI的长角血蜱遗传多样性分析

目的 了解长角血蜱的遗传多样性和不同地理种群间的系统发育关系,为长角m蜱及蜱媒病的防控奠定基础.方法 基于45条长角血蜱COI序列,采用PAUP* 4.0构建最大似然树,用DnaSP 5.0计算单倍型多样性和核苷酸多样性. 结果 长角血蜱单倍型多样性Hd为0.810~1.000,核苷酸多样性π为0.218-1.990,Fst值为0.0213-0.4522,Tajima's D为-0.7245,Fu's Fs为-2.454,分析结果均表明中国长角血蜱不同地理种群间无明显分化. 结论 系统发育分析和单倍型网络图表明长角血蜱不同地理种群正在发生扩张,但尚无明显分化.
Objective To determine the genetic diversity of Haemaphysalis longicornis and the phylogenetic relationship among different populations in order to provide a basis for the prevention and control of H.longicornis and tickborne diseases.Methods The genetic diversity of H.longicornis in China was preliminarily study based on the mitochondrial gene COL Results The results of haplotype diversity (Hd:0.810-1.000),nucleotide diversity (0.218-1.990),the Fst value (0.0213-0.4522),Tajima's D (-0.7245),and Fu's Fs (-2.454) together indicate that there was little genetic difference among different populations.Conclusion Phylogenetic analysis and haplotype networks revealed that populations of H.longicornis are expanding in different areas,but marked changes are not yet apparent.
