摘要

目的:基于高通量转录组测序及WGCNA,挖掘唾液腺多形性腺瘤(PA)恶变为癌在多形性腺瘤中(Ca-ex-PA)过程中的核心驱动基因及相关通路.方法:收集Ca-ex-PA患者肿瘤及癌旁唾液腺的新鲜标本各5例,进行高通量转录组测序(RNA-Seq)分析,筛选PA与瘤旁唾液腺组、Ca-ex-PA与癌旁唾液腺组中的差异表达基因.取PA、Ca-ex-PA共有的差异表达基因进行WGCNA分析,获得与PA恶变相关的基因群体.结果:测得良、恶性肿瘤共有差异表达基因426个,通过WGCNA得到的4个模块中,包含78个基因与恶性肿瘤表型相关.其中评分最高的EFNA3基因表达程度与高组织学分级的腺癌亚型显著相关(P<0.05).结论:WGCNA能有效筛选出与PA恶变相关的核心驱动基因,EFNA3过表达可能与PA恶变有关,其作为预后评估和治疗靶点的价值有待进一步研究.

Abstract

PURPOSE:To explore the core driving genes and related pathways of pleomorphic adenoma(PA)transforma tion into carcinoma ex pleomorphic adenoma(Ca-ex-PA)based on high-throughput transcriptome sequencing and WGCNA.METHODS:The mRNA expression of 5 cases of PA and salivary gland and 5 cases of Ca-ex-PA and salivary gland was detected by RNA-Seq.The differentially expressed genes(DEGs)of PA group and Ca-ex-PA group were screened and the intersection of DEGs of them were taken.WGCNA was used to analyze the intersection of DEGs,in order to identify the hub genes associated with malignant transformation of PA.RESULTS:Totally 426 DEGs between PA and Ca-ex-PA were detected.Through WGCNA,78 genes were considered to be highly associated with malignant transformation of PA.The highest correlation score gene was EFNA3(P<0.05).CONCLUSIONS:The malignant transformation of PA may be highly associated with EFNA3 and other hub genes.Whether EFNA3 can be used as an independent predictor in the prognosis assessment of Ca-ex-PA needs to be studied further.
