红景天甙通过NRF2/KEAP1通路对口腔鳞癌细胞生长的影响

摘要
目的:探讨红景天甙对口腔鳞癌细胞生长的影响及其机制.方法:以40 µg/mL(低剂量组)、60 µg/mL(中剂量组)、100 µg/mL(高剂量组)红景天甙处理WSU-HN30细胞,等体积培养液处理作为空白对照组.分别通过MTT法、Transwell实验及流式细胞术检测WSU-HN30细胞增殖、迁移及凋亡,通过实时定量PCR(RT-PCR)和Western免疫印迹法检测核因子E2相关因子2(NRF2)和Kelch样ECH相关蛋白1(KEAP1)mRNA和蛋白的表达.采用SPSS 20.0软件包对数据进行统计学分析.结果:同一浓度红景天甙作用WSU-HN30细胞48 h后,增殖抑制率显著高于24 h(P<0.05);同一时间,随着红景天甙浓度增加,增殖抑制率显著增加(P<0.05);与空白对照组相比,红景天甙处理后细胞迁移数显著减少,细胞凋亡率显著增加,细胞NRF2和KEAP1 mRNA及蛋白相对表达量显著降低,呈浓度依赖性(P<0.05).结论:红景天甙可显著抑制口腔鳞癌细胞增殖和迁移,诱导细胞凋亡,其机制与下调NRF2/KEAP1通路表达有关.
Abstract
PURPOSE:To investigate the effect and mechanism of salidroside on the growth of oral squamous cell carci-noma cells.METHODS:WSU-HN30 cells were divided into three salidroside treatment groups,low-dose group(40 μg/mL),middle-dose group(60 μg/mL)and high-dose group(100 μg/mL).The proliferation,migration and apoptosis of WSU-HN30 cells were detected by MTT assay,Transwell assay and flow cytometry,and the mRNA and protein expressions of nuclear-factor erythroidderived 2-like 2(NRF2)and kelch-like ECH associated protein 1(KEAP1)were detected by real-time quantitative PCR(RT-PCR)and Western blot.SPSS 20.0 software package was used for data analysis.RESULTS:At the same concentration,the proliferation inhibition rate of salidroside after 48 h was significantly higher than that after 24 h(P<0.05).At the same time,with the salidroside concentration increasing,the proliferation inhibition rate was significantly increased(P<0.05).Compared with blank control group,salidroside treatment groups had significant decrease in the num-ber of cell migration,significant increase in the rate of apoptosis,and significant decrease in mRNA and protein expres-sions of NRF2 and KEAP1;Moreover,the above changes showed a concentration-dependent manner(P<0.05).CONCLU-SIONS:Salidroside can significantly inhibit the proliferation and migration of oral squamous cell carcinoma cells,and in-duce cell apoptosis,and its mechanism may be related to the down-regulation of expression of NRF2/KEAP1 pathway.
