目的 丁香活性组分(active fraction from clove,AFC)具有明显的抗结肠癌活性,前期研究己制备AFC-PEG-PLGA纳米粒,本研究进一步系统评价AFC-PEG-PLGA纳米粒的体外、体内抗结肠癌活性.方法 常规培养人结肠癌细胞HCT116、SW620、HCT8和LOVO,以CCK-8试剂检测结肠癌细胞增殖抑制率及IC50值,筛选AFC-PEG-PLGA纳米粒敏感的结肠癌细胞;建立裸鼠HCT116结肠癌细胞移植瘤模型,随机分为阴性对照组(0.9%NS)、阳性对照组(5-氟尿嘧啶,5-Fu,30.0 mg/kg)及PEG-PLGA空纳米粒对照组、AFC(100 mg/kg)、AFC-PEG-PLGA纳米粒(100 mg/kg)治疗组.每周连续给药5 d后停药2 d,给药4周,每2~3 d测量瘤径和体重1次,计算肿瘤体积(tumor volume,TV)、相对肿瘤体积(relative tumor volume,RTV)和相对肿瘤增殖率(T/C%).实验结束后处死动物,剥取瘤组织,称瘤重,检测AFC-PEG-PLGA纳米粒对移植瘤生长的影响.结果 体外实验结果表明,AFC-PEG-PLGA纳米粒对结肠癌细胞HCT116、SW620、HCT8和LOVO的IC50为(99.58±5.48)、(145.66±6.96)、(162.62±6.58)、(229.24±9.25)μg/mL,活性最强的是HCT116细胞,活性略强于AFC.体内实验结果发现,经5-Fu、AFC与AFC-PEG-PLGA纳米粒治疗后,裸鼠TV、RTV、T/C/%及裸鼠瘤重均明显下降,其中T/C/%分别为37.54%、57.39%和43.04%,AFC-PEG-PLGA纳米粒作用强于AFC,与阴性对照组比较,P<0.01.结论 AFC-PEG-PLGA纳米粒具有较强的体外、体内抗结肠癌活性,以HCT116作用最强,活性强于AFC.
Study on the anti-colon cancer activity of the active fraction from clove PEG-PLGA nanoparticles
Objective Active fraction from clove(AFC)has significant anti-colon cancer activity.Previous studies had prepared AFC-PEG-PLGA nanoparticles,and this study explored the anti-colon cancer activity of AFC-PEG-PLGA nanoparticles in vitro and in vivo.Methods Human colon cancer cells(HCT116,SW620,HCT8 and LOVO)were routinely cultivated;colon cancer cell proliferation inhibition rate and IC50 value using the CCK-8 reagent were detected;and colon cancer cells sensitive to AFC-PEG-PLGA nanoparticles were also screened.The nude mouse model of transplanted HCT116 colon cancer cells was established and randomly divided into a negative control group(0.9%NS),a positive control group(fluorouracil,5-Fu,30.0 mg/kg),a PEG-PLGA empty nanoparticle control group,an AFC(100 mg/kg),and an AFC-PEG-PLGA nanoparticles(100 mg/kg)treatment group.After continuous administration for 5 days per week,the drug was stopped for 2 days,and the drug was administered for 4 weeks.Tumor diameter and body weight were measured every 2~3 days,and tumor volume(TV),relative tumor volume(RTV),and relative tumor proliferation rate(T/C%)were calculated.After the experiment,the animals were euthanized,tumor tissue was removed,tumor weight was measured,and the effect of AFC-PEG-PLGA nanoparticles on the growth of transplanted tumors was tested.Results In vitro experimental results showed that the IC50 of AFC-PEG-PLGA nanoparticles on colon cancer cells HCT116,SW620,HCT8,and LOVO were(99.58±5.48),(145.66±6.96),(162.62±6.58),(229.24±9.25)μg/mL,respectively.The strongest activity was observed in HCT116 cells,which were slightly stronger than AFC.In vivo experimental results showed that after treatment with 5-Fu,AFC,and AFC-PEG-PLGA nanoparticles,the TV,RTV,T/C/%,and tumor weight of nude mice were significantly reduced,with T/C/%being 37.54%,57.39%,and 43.04%,respectively.Conclusion The AFC-PEG-PLGA nanoparticles have strong anti-colon cancer activity in vitro and in vivo,with HCT116 having the strongest effect and stronger activity than AFC.
