The chemical composition identification based on UPLC-QTOF-MS/MS technology and anti-tumor,anti-inflammatory activity studies of 3 species of Paris
Objective To elucidate the bariations in the chemical,anti-tumor,and anti-inflammatory activitity of Paris polyphylla var.yunnanensis(PPY),Paris polyphylla var.chinensis(PPC)and Paris verticillata,and to serve as a guide for the rational utilization and advancement of Paris verticillata resources.Methods UPLC-QTOF-MS/MS was utilized to characterize the chemical constituents of 3 species of Paris to explore whether it contained pyrrolizidine alkaloids;the anti-tumor activity was evaluated by inhibiting the proliferation of HepG2 cells and A549 cells;and the anti-inflammatory activity was evaluated by the LPS-induced NO release model in RAW264.7 cells.Results Sixty-seven compounds were identified from the 3 species of Paris,53 of which were steroidal saponins.A total of 22 steroidal saponins were identified from Paris verticillate,and pyrrolizidine alkaloids were not identified.At the test drug concentration,Paris verticillate only inhibited the proliferation of A549 cells with an IC50 of 141.0 μg/mL,while the 3 species of Paris had no significant inhibitory effect on the release of NO from LPS-induced RAW264.7 cells at non-toxic concentrations.Conclusion Compared to PPY and PPC,Paris verticillate contained fewer types of steroidal saponins,primarily composed of polyhydroxysteroid saponins;the isolated pyrrolizidine alkaloids were not found in the samples of Paris verticillate in this experiment.In terms of pharmacological activity,Paris verticillate had potential anti-lung cancer activity.This study provided a preliminary reference for further clarifying the effective substances of Paris verticillata and the development and utilization of its resources.
