萝卜硫素通过调控OGG1基因抑制动脉粥样硬化相关巨噬细胞脂质积聚的研究

目的 探讨萝卜硫素对氧化低密度脂蛋白(ox-LDL)诱导的巨噬细胞脂质积聚的影响和作用机制.方法 将细胞分为对照组、ox-LDL组(100μg·mL-1 ox-LDL)、萝卜硫素低剂量组(100 μg·mL-1 ox-LDL+5 µmol·L-1 萝卜硫素)、萝卜硫素高剂量组(100μg·mL-1 ox-LDL±10 µmol·L-1萝卜硫素)、si-OGG1 组(100 μg·mL-1 ox-LDL+10 μmol·L-1 萝卜硫素+转染si-OGG1).以蛋白质印迹法检测各组细胞蛋白表达水平,以酶联免疫吸附测定法检测各组细胞炎性因子及8羟基脱氧鸟苷(8-OHdG)表达水平,以氧化酶法检测细胞内胆固醇水平,以探针法检测活性氧(ROS)水平,以流式细胞术检测细胞凋亡情况.结果 对照组、ox-LDL组、萝卜硫素低剂量组、萝卜硫素高剂量组、si-OGG1组的8-氧代鸟嘌呤DNA糖基化酶1(OGG1)蛋白相对表达水平分别为 0.80±0.03、0.26±0.02、0.43±0.03、0.68±0.05 和 0.29±0.04,8-OHdG水平分别为(3.02±0.16)、(23.02±1.07)、(18.28±1.40)、(13.05±1.42)和(17.39±1.53)ng·mL-1,总胆固醇含量分别为(0.08±0.01)、(0.39±0.01)、(0.28±0.01)、(0.17±0.02)和(0.30±0.02)mmol·g-1,ROS 水平分别为 1.66±0.10、8.23±0.78、5.41±0.15、3.27±0.24和 7.01±0.41,细胞凋亡率分别为(3.64±0.31)%、(21.72±2.70)%、(16.60±1.18)%、(10.04±0.52)%和(16.07±1.46)%.对照组的上述指标与ox-LDL组比较,萝卜硫素低、高剂量组的上述指标与ox-LDL组比较,si-OGG1组的上述指标与萝卜硫素高剂量组比较,在统计学上差异均有统计学意义(均P<0.05).结论 萝卜硫素可能通过上调OGG1表达抑制ox-LDL诱导的巨噬细胞脂质积聚.
Sulforaphane inhibits lipid accumulation in atherosclerotic macrophages by regulating OGG1 gene
Objective To investigate the effect of sulforaphane on lipid accumulation in macrophages induced by oxidized low-density lipoprotein(ox-LDL)and its mechanism.Methods The cells were divided into control group,ox-LDL group(100 µg·mL-1 ox-LDL),sulforaphane-L group(100 µg·mL-1 ox-LDL+5 µg·mL-1 sulforaphane),sulforaphane-H group(100 µg·mL-1 ox-LDL+10 µg·mL-1 sulforaphane)and si-OGG1 group(100 µg·mL-1 ox-LDL+10 μg·mL-1 sulforaphane+transfected with si-OGG1).Western blot assay was used to detect the expression level of protein in each group,enzyme linked immunosorbent assay was used to detect the expression of inflammatory factors and 8-hydroxydeoxyguanosine(8-OHdG)in each group,oxidase assay was used to detect intracellular cholesterol level,and reactive oxygen species(ROS)level was detected by probe method,apoptosis was detected by flow cytometry.Results The OGG1 protein expression levels of control group,ox-LDL group,sulforaphane-L group,sulforaphane-H group and si-OGG1 group were 0.80±0.03,0.26±0.02,0.43±0.03,0.68±0.05 and 0.29±0.04;the 8-OHdG levels were(3.02±0.16),(23.02±1.07),(18.28±1.40),(13.05±1.42)and(17.39±1.53)ng·mL-1;total cholesterol content were(0.08±0.01),(0.39±0.01),(0.28±0.01),(0.17±0.02)and(0.30±0.02)mmol·g-1;ROS levels were 1.66±0.10,8.23±0.78,5.41±0.15,3.27±0.24 and 7.01±0.41;the apoptosis rates were(3.64±0.31)%,(21.72±2.70)%,(16.60±1.18)%,(10.04±0.52)%and(16.07±1.46)%,respectively.Compared between control group and ox-LDL group,compared between ox-LDL group and sulforaphane-L,sulforaphane-H groups,compared between si-OGG1 group and sulforaphane-H group,the differences of the above indexes were all statistically significant(all P<0.05).Conclusion Sulforaphane may inhibit ox-LDL-induced lipid accumulation in macrophages by up-regulating OGG1 expression.
