摘要
共药(codrug)是一类以2个或多个具有药理活性的药物分子以共价结合的方式形成的前药型化合物,在体内经过活化释放出原药发挥治疗作用.这种药物设计具有协同药效、降低药物不良反应发生率、改善稳定性和药代动力学特征、定向释放等特点,但随着制药技术的发展,这些特点逐渐被其他设计所替代.本文将通过对共药的特点及应用进行介绍,浅析此类药物在药物开发策略中的利弊和展望.
Abstract
Codrug is a kind of prodrug compound formed by covalent combination of two or more pharmacologically active drug molecules,which is activated in vivo to release the original drug to play a therapeutic effect.This kind of drug design has the characteristics of synergistic efficacy,reducing incidence of adverse drug reactions,improving stability and pharmacokinetic characteristics,targeted release,etc.,but with the development of pharmaceutical technology,these characteristics are gradually replaced by other designs.This article will introduce the characteristics and application of co-drugs,and analyze the advantages,disadvantages and prospects of such drugs in drug development strategy.
